I did the better part of the GAP last week myself. It was awesome, though not as colorful as I'd expected honestly. I think this past week or right now is the time to be there for peak viewing.

i would start in ohiopyle or confluence, the one day trip between confluence and cumberland is the best daytrip the trail has to offer

I concur with Mark.
I think the trail form Connelsville to Confluence is the most picturesque of the trip, with continuing on over the divide and into Cumberland being a pretty amazing day on the bike.

Connellsville to Confluence was nice. Decent number on the trail and in Sisters Cafe - Pumpkin Milkshake was worth the trip.
